Operating Procedure SOP

The Olympus OLS 4000 LEXT is a laser confocal microscope that can take high resolution 3D images by acquiring successive images of the sample between two heights. It then recombines the laser acquired images and the color images to produce a 3D projection that can be measured. It can measure dimensions, roughness, area, volumes, etc. Its specially developed lenses produce high quality images of the sample. The z axis resolution is 10 nm and the x-y resolution is 120 nm.

Please follow the steps below when finishing your work:

  • Send the stage to LOAD to remove your sample
  • Remove sample, send stage back
  • Exit the LEXT program
  • Turn off the LEXT power supply

Do not forget to transfer your data to the LNF Network from the Temporary file where you saved your data

The results of a recent maintenance/calibration on the LEXT OLS4000 are shown below. The height measurement have the following error:

20X 0.793%

50X 1.061%

100X 0.213%

The stitching was tested using one of the standards: on a 2x2 stitch using 50 X lens

  • Coarse stage adjustment allows examination of tall (10 cm ) samples

Substrate requirements

  • Max sample diameter 6"
  • Maximum sample thickness 10 cm
  • Maximum sample weight 1 kg
  • Small samples should be placed on a glass slide to avoid cross-contamination

Material restrictions

This microscope can be used with any material as long as it weighs less than 1.5 kg and fits comfortably under the lenses.

The Olympus OLS 4000 LEXT is designated as a General class tool. Below is a list of approved materials for the tool. Approved means the material is allowed in the tool under normal circumstances. If a material is not listed, please create a helpdesk ticket or email info@lnf.umich.edu for any material requests or questions.

Checkout procedure

  1. Read through this page and the Standard Operating Procedure above.
  2. Create a Helpdesk Ticket requesting training.
  3. A tool engineer will schedule a time for initial training.
  4. Practice with your mentor or another authorized user until you are comfortable with tool operation.
  5. Schedule a checkout session with a tool engineer via the Helpdesk ticket system. If this checkout is successful, the engineer will authorize you on the tool.


Maintenance for this tool is done regularly using height standards.
